چکیده
Abstract Background Anti-PD-1/PD-L1 immunotherapy is a promising treatment for ESCC. High PD-L1 expression theoretical predictive biomarker, but the poor response also observed in some patients with high expression. Biopsies assessment of always focuses on single region which may be influenced by spatial heterogeneity tumor microenvironment. This study investigates whether biopsy different regions related to its paradoxical value. Methods We prospectively included 30 consecutive treatment-naïve, stage II-III ESCC who underwent upfront esophagectomy. Multi-region sampling was taken endoscopic forceps (Fig. A) from four (A/B/C/D) proximal, distal, midpoint surface, and internal center, respectively. Each spaced ≥1 cm apart avoided areas severe necrosis. compared at endoscopic-biopsy overall surgical-resected specimen. evaluated immunohistochemistry using combined positive score (CPS). negative defined as CPS ≥ 1 < 1, Results Based tumor, were 66.7% 10% patients, positive/negative discordance rate concordance (kappa = 0.206) B). Correlations between C) revealed B D (r 0.77) relatively low others. Combining multi-region evaluation improved correlation consistency D). Comparing based gold standard specimen, sensitivity increased 12.5% versus single-region assessment. Conclusion spatially heterogeneous ESCC, cannot reflect status consistent observation suboptimal prediction immunotherapy. could help improve bias heterogeneity, identification candidate

Prognostic significance of tumor-infiltrating immune cells and PD-L1 expression in esophageal squamous cell carcinoma
Programmed death-1 receptor (PD-1) and its ligand (PD-L1) play an integral role in regulating the immune response against cancer. This study investigated the prognostic significance of PD-L1 expression on tumor cells and tumor-infiltrating immune cells (TILs) in the tumor microenvironment in Chinese patients with esophageal squamous cell carcinoma (ESCC). Archival formalin-fixed, paraffin-embed...
متن کاملIndependent prognostic role of PD-L1 expression in patients with esophageal squamous cell carcinoma
Accumulating evidence has shown that PD-L1 expression is associated with clinicopathological features in various human malignancies. We searched for correlations between PD-L1 expression and clinicopathological data in esophageal squamous cell carcinoma (ESCC) patients. PD-L1 expression in primary tumors from 278 patients was evaluated using immunohistochemistry (IHC) in ESCC tissue microarray....
متن کاملCorrection: Elevated Maspin Expression Is Associated with Better Overall Survival in Esophageal Squamous Cell Carcinoma (ESCC)
Tumor suppressor maspin is a differentially regulated gene in the progression of many types of cancer. While the biological function of maspin in blocking tumor invasion and metastasis is consistent with the loss of maspin expression at the late stage of tumor progression, the differential expression and the biological significance of maspin in early stage of tumor progression appear to be comp...
متن کاملPrognostic value of PD-L1 in esophageal squamous cell carcinoma: a meta-analysis
Accumulated evidence has shown that the programmed cell death receptor 1/programmed cell death receptor 1 ligand 1 (PD-1/PD-L1) pathway is a promising therapeutic target for cancer immunotherapy. However, the association between PD-L1 and esophageal squamous cell carcinoma (ESCC) patient survival remains unclear. We performed a meta-analysis to investigate the prognostic value of PD-L1 in ESCC....
متن کاملExpression of ubiquitin C-terminal hydrolase L1 (UCH-L1) and its prognostic significance in esophageal squamous cell carcinoma
Objective: To explore the expression of ubiquitin carboxyl-terminal esterase L1 (UCH-L1) in the esophageal squamous cell carcinoma (ESCC) as well as its clinical significance. Methods: 215 cases of ESCC were evaluated by tissue microarray and immunohistochemistry for UCH-L1 expression and its clinical significance and prognostic value was investigated. Results: The positive rate of UCH-L1 was 4...
